Breakthrough Nasal Spray Shows Promise in Reversing Brain Aging

As the human brain ages, it doesn’t simply wear out — it often falls victim to a slow-burning buildup of inflammation, according to the latest research. This chronic irritation, known as “neuroinflammaging,” occurs deep within the brain’s memory centers. For decades, the resulting brain fog and increased risk of neurodegenerative diseases like Alzheimer’s and dementia were considered an inevitable part of growing older. Now, groundbreaking research from Texas A&M University suggests this process is not only treatable but entirely reversible.A team at the university’s College of Medicine has developed an innovative nasal spray that targets this age-related cognitive decline. Led by Dr. Ashok Shetty, alongside senior research scientists Dr. Madhu Leelavathi Narayana and Dr. Maheedhar Kodali, the study recently published in the “Journal of Extracellular Vesicles” details how just two doses of the therapy dramatically reduced inflammation and improved memory in subjects.“Brain age-related diseases like dementia are a major health concern worldwide,” Dr. Shetty said in a press release. “What we’re showing is brain aging can be reversed, to help people stay mentally sharp, socially engaged, and free from age-related decline.”The therapy relies on extracellular vesicles, which act as microscopic biological delivery vehicles. These EVs are loaded with microRNAs, powerful genetic cargo that regulates brain functions.“MicroRNAs act like master regulators,” Dr. Narayana said. “They help modulate and regulate many gene and signaling pathways in the brain.” By using a nasal spray, researchers can bypass the brain’s protective barriers, allowing the EVs to travel directly into the brain tissue.“The mode of delivery is one of the most exciting aspects of our approach,” Dr. Kodali said. “Intranasal delivery allows us to reach, and treat, the brain directly without invasive procedures.”Once absorbed, the microRNAs suppress inflammatory pathways. Furthermore, the therapy successfully recharges the brain’s mitochondria — the power plants of the cells. “We are giving neurons their spark back by reducing oxidative stress and reactivating the brain’s mitochondria,” Dr. Narayana said.Following treatment, behavioral tests showed remarkable improvements in object recognition and spatial awareness. The results appeared within weeks and were sustained for months. “We are seeing the brain’s own repair systems switch on, healing inflammation and restoring itself,” Dr. Shetty said.Crucially, the therapy proved equally effective in both males and females, a rare and highly sought-after outcome in biomedical studies. “It’s universal,” Dr. Shetty said. “Treatment outcomes were consistent and similar across both sexes.”The urgency for such therapies is mounting. In the United States, new dementia cases are projected to rise from just over 500,000 in 2020 to about 1 million by 2060. “The trend signals a pressing need for policies and innovative interventions that can minimize both the risk and severity of neurodegenerative disorders like dementia,” Dr. Shetty said.If successfully scaled, this new therapy could radically alter the landscape of aging, replacing heavy medications with a simple, non-invasive spray. “Our approach redefines what it means to grow old. We’re aiming for successful brain aging: keeping people engaged, alert, and connected. Not just living longer, but living smarter and healthier,” the doctor said.
