World’s First Human Trial Begins for Revolutionary Tooth-Regrowing Drug

Japanese researchers have officially begun the world’s first human clinical trials for a groundbreaking experimental drug designed to regrow teeth, hoping to completely revolutionize modern dentistry and offer hope to millions who suffer from tooth loss due to congenital conditions, decay, or injury.The experimental biologic, known as TRG-035, was developed by Toregem BioPharma, a biotech startup spun out of Kyoto University. The drug works by targeting a specific protein called USAG-1, which functions as a molecular parking brake that permanently shuts down tooth formation after our adult teeth emerge. By neutralizing this biological barrier with an antibody injection, the drug reactivates dormant tooth buds that remain in the human jaw. The drug effectively blocks that protein, which the researchers call “the body’s molecular off-switch for tooth development.”The discovery initially stemmed from a genetic accident. Researchers found that mice bred without the gene for that protein sprouted teeth they were never supposed to have. Following this realization, scientists successfully tested the treatment on animals. One shot of the antibody later produced working teeth in mice and ferrets, with no serious safety problems.Now, the focus has shifted to humans. The Phase I clinical trial at Kyoto University Hospital is focuses strictly on human safety. The initial study enrolled 30 adult men, aged 30 to 64, who are each missing at least one tooth. The first round, which began in late 2024 and was tracked for 11 months, tests only whether the drug is safe in people. Because of this, the researchers say “actual tooth regrowth in these adult participants is unlikely.”To push the research to its next crucial stage, Toregem BioPharma recently raised $5.3 million, bringing the company’s total funding past the $29 million mark. This substantial financial backing will support Phase II clinical trials. If the drug clears the safety stage, the next round will treat children aged 2 to 7 who did not develop teeth during prenatal gestation. About one person in a thousand is born missing six or more, which makes chewing hard and, the researchers say, prompts enough cruel teasing that some teenagers wear masks. Toregem BioPharma wants the drug on the market by 2030.“We want to do something to help those who are suffering from tooth loss or absence,” lead researcher Katsu Takahashi, head of dentistry and oral surgery at Kitano Hospital, said before the Phase 1 trial began. “While there has been no treatment to date providing a permanent cure, we feel that people’s expectations for tooth growth are high.”
